Overview

Canocapavir (JNJ-6379) is a small molecule capsid assembly modulator (CAM) that was under development by Janssen for the treatment of chronic hepatitis B virus (HBV) infection. It functions by binding to the HBV core protein, inducing the assembly of aberrant or empty capsids, thereby preventing the encapsidation of pregenomic RNA and subsequent viral replication. In clinical development, canocapavir was frequently studied in combination with nucleos(t)ide analogs (NUCs), such as tenofovir or entecavir, which act as reverse transcriptase inhibitors to suppress viral DNA synthesis. This combination approach was intended to target multiple stages of the HBV lifecycle to improve functional cure rates. Janssen discontinued the development of canocapavir in late 2022 as part of a strategic reprioritization of its HBV portfolio.
