Overview

Canosimibe + atorvastatin is a combination therapy developed for the treatment of primary hypercholesterolemia, particularly in patients inadequately controlled on statin therapy alone. Canosimibe (AVE5530) is an inhibitor of Niemann-Pick C1-like protein 1 (NPC1L1), which reduces intestinal cholesterol absorption[4]. Atorvastatin is an HMG-CoA reductase inhibitor (statin) that decreases hepatic cholesterol synthesis. The combination was investigated to provide additive or synergistic reduction in LDL-C and other lipid parameters by targeting two complementary pathways for cholesterol lowering. Clinical trials in patients with primary hypercholesterolemia demonstrated efficacy of the combination in lowering LDL-C but development was discontinued after reaching phase 3 trials[1][3][4][6].
