Overview

Capecitabine + carboplatin is a combination chemotherapy regimen consisting of the oral fluoropyrimidine carbamate capecitabine and the intravenous platinum-based agent carboplatin. Capecitabine acts as a prodrug that is enzymatically converted to 5-fluorouracil (5-FU) within the body, primarily in tumor tissue, where it inhibits thymidylate synthase and interferes with DNA synthesis. Carboplatin is a second-generation platinum compound that forms reactive platinum complexes which bind to DNA, creating intra-strand and inter-strand cross-links that inhibit DNA replication and transcription, ultimately leading to apoptosis. This combination is utilized across several oncology indications, including esophagogastric, breast, and ovarian cancers, often chosen for its favorable toxicity profile—specifically lower neurotoxicity compared to oxaliplatin-based regimens—and ease of administration.
