Overview

This is a combination regimen consisting of three agents used in oncology: - **Capecitabine** is an oral prodrug of 5-fluorouracil (5-FU), classified as an antimetabolite. It interferes with DNA synthesis by inhibiting thymidylate synthase, leading to impaired cell division and tumor growth suppression[9]. - **Cetuximab** is a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR/ErbB1). By binding to EGFR, it blocks ligand-induced activation and downstream signaling pathways involved in cell proliferation and survival[1]. - **Lapatinib** is a small molecule tyrosine kinase inhibitor that targets both EGFR (ErbB1) and HER2 (ErbB2) receptors. It inhibits their phosphorylation, thereby blocking downstream PI3K/AKT and MAPK signaling pathways critical for cancer cell growth[1]. The rationale for combining these agents lies in their complementary mechanisms—capecitabine provides cytotoxic chemotherapy, while cetuximab and lapatinib offer dual blockade of the ErbB family receptors at different levels (extracellular via antibody; intracellular via kinase inhibition)[1]. This combination has been explored primarily in solid tumors such as colorectal cancer or head and neck cancers where EGFR/HER2 pathways are implicated.
