Overview

This is a four-drug combination chemotherapy regimen consisting of capecitabine, cyclophosphamide, docetaxel, and doxorubicin. Each component has a distinct mechanism of action targeting cancer cell proliferation and survival: - **Capecitabine** is an oral prodrug that is converted to 5-fluorouracil (5-FU) in the body. It acts as an antimetabolite by inhibiting thymidylate synthase, thereby interfering with DNA synthesis and repair in rapidly dividing cells. - **Cyclophosphamide** is an alkylating agent that crosslinks DNA strands, leading to inhibition of DNA replication and cell death. - **Docetaxel** is a taxane that stabilizes microtubules and prevents their depolymerization during mitosis, resulting in cell cycle arrest and apoptosis. - **Doxorubicin** is an anthracycline antibiotic that intercalates into DNA, inhibits topoisomerase II activity (preventing proper DNA replication), and generates free radicals causing cytotoxicity. This combination targets multiple pathways involved in cancer cell growth. It has been investigated primarily for the treatment of breast cancer—especially high-risk or node-positive early-stage or metastatic disease—where multi-agent regimens are used to maximize tumor response[2][4][7]. The rationale for combining these agents includes their complementary mechanisms of action as well as evidence for synergistic antitumor effects[5].
