Overview

Capecitabine + nimotuzumab is an investigational combination therapy consisting of capecitabine, an oral prodrug of 5-fluorouracil (5-FU), and nimotuzumab, a humanized monoclonal antibody targeting the epidermal growth factor receptor (EGFR). Capecitabine acts as a small molecule antimetabolite that inhibits DNA synthesis in rapidly dividing tumor cells. Nimotuzumab binds to EGFR on the surface of cancer cells, blocking ligand-induced activation and downstream signaling pathways involved in cell proliferation and survival. This combination is being studied primarily for its potential to improve outcomes in patients with recurrent or metastatic nasopharyngeal carcinoma who have failed first-line platinum-based chemotherapy. The rationale for combining these agents is based on their complementary mechanisms—capecitabine’s cytotoxic effects and nimotuzumab’s targeted inhibition of EGFR-mediated tumor growth[1][2][10]. Clinical trials are ongoing to evaluate efficacy and safety.
