This is a four-drug combination regimen used primarily in the treatment of metastatic colorectal cancer. It consists of capecitabine (an oral prodrug of 5-fluorouracil), oxaliplatin (a platinum-based chemotherapeutic), bevacizumab (a monoclonal antibody targeting vascular endothelial growth factor A, VEGF-A), and cetuximab (a monoclonal antibody targeting epidermal growth factor receptor, EGFR). Capecitabine and oxaliplatin act as cytotoxic agents interfering with DNA synthesis and function. Bevacizumab inhibits angiogenesis by blocking VEGF-A, thereby reducing tumor blood supply. Cetuximab blocks EGFR-mediated signaling pathways involved in cell proliferation and survival. This combination has been studied as first-line therapy for advanced or metastatic colorectal cancer; however, clinical trials have shown that adding cetuximab to the standard XELOX-bevacizumab regimen does not improve efficacy but increases toxicity[1][6]. The individual drugs are developed by different companies.
