Overview

This treatment regimen is a standard-of-care chemoradiotherapy protocol used as a comparator in clinical trials for locally advanced rectal cancer at the Hebei Medical University Fourth Hospital. It consists of long-course radiotherapy (LCRT) administered concurrently with capecitabine, followed by consolidation chemotherapy using the CAPOX regimen (capecitabine and oxaliplatin). Capecitabine is an oral fluoropyrimidine carbamate that acts as a cytotoxic antimetabolite by inhibiting thymidylate synthase, while oxaliplatin is a platinum-based antineoplastic agent that forms DNA cross-links. Radiotherapy uses ionizing radiation to induce DNA damage in tumor cells. This specific protocol is being evaluated against total neoadjuvant therapy (TNT) involving short-course radiotherapy and immunotherapy.
