Overview

Capecitabine + toripalimab is a combination therapy being investigated as a maintenance treatment for patients with recurrent or metastatic nasopharyngeal carcinoma (NPC). Toripalimab is a humanized IgG4-κ monoclonal antibody that acts as an immune checkpoint inhibitor by targeting the programmed cell death protein 1 (PD-1) receptor on T cells, thereby restoring anti-tumor immune responses. Capecitabine is an orally administered fluoropyrimidine carbamate prodrug that is enzymatically converted to 5-fluorouracil (5-FU) within tumors. 5-FU inhibits thymidylate synthase, interfering with DNA synthesis and cell division. This combination is specifically being evaluated in a Phase 3 clinical trial (NCT05233358) led by Yan-Qun Xiang at Sun Yat-sen University Cancer Center to improve progression-free survival in NPC patients who have achieved disease control following first-line induction chemotherapy.
