Overview

Capecitabine + tucatinib is a combination of two oral small molecule drugs used primarily in the treatment of HER2-positive metastatic breast cancer. Capecitabine is an antimetabolite chemotherapy agent that is converted in the body to 5-fluorouracil (5-FU), which inhibits DNA synthesis and cell division. Tucatinib is a highly selective tyrosine kinase inhibitor targeting the human epidermal growth factor receptor 2 (HER2), blocking downstream signaling pathways involved in tumor cell proliferation and survival. This combination is often administered with trastuzumab for patients who have received prior anti-HER2 therapies, including those with brain metastases[2][4][5]. The regimen has demonstrated improved progression-free survival and overall survival compared to standard therapy.
