Capivasertib + docetaxel is a combination therapy under investigation primarily for metastatic castration-resistant prostate cancer (mCRPC) and other solid tumors. Capivasertib is an orally bioavailable small molecule inhibitor of all three isoforms of the serine/threonine kinase AKT (AKT1, AKT2, and AKT3), a key component of the PI3K/AKT signaling pathway often dysregulated in cancer. Docetaxel is a taxoid antineoplastic agent that promotes microtubule assembly and stability, inhibiting cell division by preventing microtubule depolymerization. The combination aims to enhance anti-tumor activity by targeting both proliferating tumor cells (via docetaxel) and residual persister cells with activated survival pathways (via capivasertib). Preclinical studies show that this combination reduces proliferation and increases apoptosis in prostate cancer models regardless of PTEN status[1][4][5]. Clinical trials are ongoing to evaluate efficacy and safety in mCRPC[3][6].
