Overview
Capivasertib + fulvestrant is a fixed combination regimen of two drugs used for the treatment of hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative locally advanced or metastatic breast cancer in adults whose tumors have one or more PIK3CA/AKT1/PTEN alterations. Capivasertib is an oral small molecule inhibitor targeting AKT kinases (a serine/threonine kinase in the PI3K/AKT pathway), which blocks abnormal signaling that drives cancer cell proliferation and survival. Fulvestrant is an estrogen receptor antagonist and degrader that binds to and accelerates degradation of the estrogen receptor, thereby inhibiting estrogen-driven tumor growth. The combination was shown to significantly improve progression-free survival compared to fulvestrant alone in patients with relevant biomarker alterations[1][2][4][5][9].
