Capivasertib + olaparib is a novel combination therapy that targets two fundamental weaknesses in cancer simultaneously. Olaparib is a poly ADP ribose polymerase (PARP) inhibitor that targets DNA damage repair mechanisms, while capivasertib is an AKT inhibitor that blocks the PI3K/AKT pathway which fuels tumor growth. This dual-pronged approach has shown promising antitumor activity in various advanced solid tumors, particularly in patients with germline BRCA1/2 mutations and BRCA1/2 wild-type cancers. The combination has demonstrated durable activity in ovarian, endometrial, and breast cancers, with especially promising responses observed in endometrial cancer.
