CAR-HSC (EGFRvIII) is an experimental hematopoietic stem cell (HSC)-based chimeric antigen receptor (CAR) therapy being developed by Immugenia for the treatment of glioblastoma (GBM). The therapy involves engineering hematopoietic stem cells to express a CAR directed against EGFRvIII (epidermal growth factor receptor variant III), a tumor-specific mutation found in approximately 30-50% of EGFR-amplified glioblastomas. Unlike conventional CAR-T therapies that utilize mature T cells, this platform uses engineered HSCs designed to continuously generate both CAR-T and CAR-NK cells in vivo. This stem cell-based approach aims to provide sustained anti-tumor immunity, improved persistence, and potentially reduced toxicity compared to traditional adoptive cell transfer methods. The program is currently in the discovery stage of development.
