Overview

CAR-M (Cancer Institute and Hospital, Chinese Academy of Medical Sciences) is an investigational chimeric antigen receptor macrophage (CAR-M) cell therapy being developed for the treatment of advanced solid tumors. Utilizing an adenovirus vector platform, macrophages are engineered to express CARs targeting specific tumor-associated antigens, including HER2, PSMA, and FAP. Unlike traditional CAR-T cell therapies, CAR-M cells are designed to better infiltrate the dense, immunosuppressive tumor microenvironment of solid tumors. Once at the tumor site, these engineered macrophages can directly phagocytose tumor cells and secrete pro-inflammatory cytokines to remodel the microenvironment and present tumor antigens to endogenous T cells, thereby stimulating a broader adaptive immune response. The therapy is currently being evaluated in a Phase I platform clinical trial to assess safety, tolerability, and preliminary efficacy across multiple target-specific cohorts.
