CAR NK cells (non-edited) refer to an experimental chimeric antigen receptor (CAR) natural killer (NK) cell therapy that has not undergone additional genomic modifications beyond the insertion of the CAR construct. In the research context presented at ASH 2024, these cells are derived from umbilical cord blood (CB) and engineered to express a CAR targeting B-cell maturation antigen (BCMA), typically incorporating a 4-1BB costimulatory domain. These non-edited cells serve as the baseline therapeutic product in studies evaluating the impact of CRISPR/Cas9-mediated multiplex editing (targeting genes like KLRC1, TGFBR2, and PRDM1) to overcome immunosuppression and improve in vivo persistence. Preclinical data in multiple myeloma models indicate that while these cells possess significant antitumoral activity, their efficacy and proliferative capacity are further enhanced by subsequent gene editing.
