CD19-targeted CAR-T cell therapy is a revolutionary class of immunotherapy where a patient's own T cells (autologous) or donor T cells (allogeneic) are genetically modified to express a chimeric antigen receptor (CAR) that recognizes the CD19 protein. CD19 is a biomarker found on the surface of nearly all B-lineage cells, including those in B-cell leukemias and lymphomas. Once infused back into the patient, these CAR-T cells identify and bind to CD19-positive cells, triggering T-cell activation and the subsequent destruction of the target B cells. This approach has led to significant clinical success and regulatory approvals for several products in treating relapsed or refractory B-cell malignancies such as acute lymphoblastic leukemia (ALL), diffuse large B-cell lymphoma (DLBCL), and mantle cell lymphoma (MCL). Beyond oncology, CD19 CAR-T therapies are being investigated for their potential to treat severe B-cell-mediated autoimmune diseases like systemic lupus erythematosus (SLE) by depleting the pathogenic B-cell population.
