CAR-T-BCMA + PD-1 refers to a combination therapy involving chimeric antigen receptor T-cell (CAR-T) therapy targeting B-cell maturation antigen (BCMA) and a programmed cell death protein 1 (PD-1) inhibitor. CAR-T therapies are autologous or allogeneic T cells genetically engineered to express a synthetic receptor that recognizes BCMA, an antigen highly expressed on malignant plasma cells in multiple myeloma. These modified T cells can directly target and kill BCMA-expressing cancer cells[2][3]. The addition of a PD-1 inhibitor aims to overcome immune exhaustion and enhance the persistence and efficacy of CAR-T cells by blocking the inhibitory PD-1 pathway, which tumors often exploit to evade immune attack. Preclinical data suggest that combining BCMA-targeted CAR-T with anti-PD-1 agents may improve cytotoxic function against myeloma, especially in cases where relapse occurs after initial CAR-T therapy[2]. Clinical trials are ongoing to evaluate this combination approach in relapsed/refractory multiple myeloma.
