Drug intelligence / Profile preview
CD19-targeted chimeric antigen receptor (CAR) T-cell therapy is a class of cellular immunotherapies designed to target and eliminate B cells expressing the CD19 antigen. This therapy involves harvesting a patient's (or donor's) T cells, genetically engineering them ex vivo to express a CAR specific to CD19, expanding them, and reinfusing them into the patient. CD19 is a transmembrane glycoprotein widely expressed across B-cell development stages and on most B-cell malignancies, making it a highly effective target for hematologic cancers. Several CD19-targeted CAR T-cell therapies have been approved by regulatory agencies, including tisagenlecleucel (Kymriah), axicabtagene ciloleucel (Yescarta), brexucabtagene autoleucel (Tecartus), and lisocabtagene maraleucel (Breyanzi), primarily for the treatment of relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL), diffuse large B-cell lymphoma (DLBCL), and other B-cell lymphomas. Additionally, CD19 CAR-T therapies are being actively investigated for severe autoimmune diseases such as systemic lupus erythematosus (SLE) and systemic sclerosis.
