Overview

CAR T-cell therapy (CD19-targeting) is a class of adoptive cell immunotherapies in which a patient's T cells are genetically engineered to express chimeric antigen receptors (CARs) that specifically recognize and bind to CD19, a surface glycoprotein expressed on B cells and most B-cell malignancies. Upon binding to CD19, the CAR T cells are activated to proliferate and eliminate the target cells through cytotoxic mechanisms. This therapeutic approach has been highly successful in treating relapsed or refractory B-cell hematologic malignancies, including acute lymphoblastic leukemia (ALL), diffuse large B-cell lymphoma (DLBCL), and mantle cell lymphoma (MCL). Several products have received regulatory approval, such as tisagenlecleucel (Kymriah), axicabtagene ciloleucel (Yescarta), and lisocabtagene maraleucel (Breyanzi). Additionally, CD19-targeting CAR T-cell therapy is being explored for the treatment of severe autoimmune diseases, including systemic lupus erythematosus (SLE) and chronic inflammatory demyelinating polyradiculoneuropathy (CIDP), where depleting pathogenic B cells may provide therapeutic benefit.
