This experimental CAR-T cell therapy is an investigational cell-based immunotherapy being developed by Guangzhou 8th People's Hospital in collaboration with Sun Yat-sen University for the treatment of HIV-1 infection. The therapy involves T cells genetically engineered to express chimeric antigen receptors (CARs) derived from broadly neutralizing antibodies (bNAbs) that specifically target the HIV-1 envelope glycoprotein (Env) on the surface of infected cells. It is currently being evaluated in a Phase 1 clinical trial (NCT03980691) as part of a 'shock and kill' strategy. In this approach, the CAR-T cells are administered in combination with Chidamide, a histone deacetylase (HDAC) inhibitor that acts as a latency-reversing agent to reactivate dormant HIV-1 from the latent reservoir, allowing the engineered immune cells to identify and eliminate the reactivated infected cells in patients receiving suppressive combination antiretroviral therapy (cART).
