Drug intelligence / Profile preview
CAR-T cells (B7-H3-targeting) refers to chimeric antigen receptor (CAR) T-cell therapies engineered to target B7-H3 (CD276), an immunoregulatory transmembrane glycoprotein overexpressed in various solid tumors and central nervous system (CNS) malignancies. These therapies involve genetically modifying autologous or allogeneic T cells to express a CAR that specifically binds to B7-H3, enabling them to recognize and eliminate B7-H3-expressing cancer cells. B7-H3 is highly expressed in pediatric solid tumors, brain tumors (such as diffuse intrinsic pontine glioma [DIPG], diffuse midline glioma [DMG], and diffuse hemispheric glioma [DHG]), pancreatic cancer, and adrenocortical carcinoma (ACC), while showing limited expression in normal tissues. Multiple clinical trials, such as the BrainChild-03 trial, are evaluating B7-H3-targeting CAR-T cells, demonstrating safety, feasibility, and potential efficacy in early-phase studies.
