CAR19 (John Lister) is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting the CD19 antigen, developed for the treatment of various B-cell malignancies including B-cell lymphoma and B-cell acute lymphoblastic leukemia. This therapy is notable for its decentralized, "point-of-care" manufacturing approach, which utilizes the Miltenyi Biotec CliniMACS Prodigy automated cell processing system. T-cells are harvested from the patient and transduced with a lentiviral vector (CAR19) provided by Lentigen, a subsidiary of Miltenyi Biotec. The automated system handles the cell selection, transduction, and expansion in a closed environment, allowing for local manufacture at the clinical site. Once the CAR T-cells are generated, they are re-infused into the patient to target and eliminate CD19-expressing B-cells. This model aims to reduce turnaround times and logistical complexities associated with centralized CAR-T production facilities.
