Overview

This combination therapy consists of eribulin mesylate (a microtubule dynamics inhibitor), carboplatin (a platinum-based DNA alkylating agent), and veliparib (a PARP inhibitor). The regimen is being investigated by The University of Texas Health Science Center at San Antonio in a Phase II clinical trial for patients with Stage IV (metastatic) breast or ovarian cancer who exhibit homologous recombination deficiency (HRD) or BRCA mutations. Eribulin inhibits the growth phase of microtubules without affecting the shortening phase, leading to cell cycle arrest. Carboplatin creates DNA crosslinks, and veliparib prevents the repair of these DNA lesions by inhibiting poly(ADP-ribose) polymerase (PARP) enzymes, potentially creating a synergistic effect in HRD-positive tumors.
