This combination regimen is a multi-agent therapeutic approach being evaluated as a standard-of-care comparator in clinical trials for extensive-stage small cell lung cancer (ES-SCLC). The regimen consists of an induction phase combining carboplatin (a platinum-based DNA alkylating agent), etoposide (a topoisomerase II inhibitor), and atezolizumab (a PD-L1-targeting monoclonal antibody), followed by a maintenance phase with atezolizumab and lurbinectedin (a selective inhibitor of oncogenic transcription). By combining cytotoxic chemotherapy, immune checkpoint inhibition, and a novel transcription inhibitor, the regimen aims to provide a comprehensive treatment strategy for patients with advanced SCLC. The individual components are developed by various companies, including Roche (atezolizumab) and PharmaMar/Jazz Pharmaceuticals (lurbinectedin), and are being utilized in Phase 3 trials sponsored by SystImmune.
