Overview

This is a combination chemotherapy regimen consisting of three cytotoxic agents—carboplatin, fluorouracil (5-fluorouracil or 5-FU), and irinotecan. Each drug has a distinct mechanism of action targeting rapidly dividing cancer cells: - **Carboplatin** is an organoplatinum alkylating agent that forms DNA crosslinks, inhibiting DNA synthesis and function, leading to cell death[6]. - **Fluorouracil** is an antimetabolite that inhibits thymidylate synthase and incorporates into RNA and DNA, disrupting their synthesis and function[5]. - **Irinotecan** is a topoisomerase I inhibitor; it prevents the religation of single-strand breaks in DNA during replication, resulting in lethal double-strand breaks[2][3]. This combination may be used for advanced or metastatic cancers such as colorectal cancer when other regimens are not suitable or as part of clinical trials. The drugs are administered intravenously.
