Overview

This is a combination regimen consisting of three drugs: - **Carboplatin** is a platinum-based chemotherapeutic agent that causes DNA crosslinking and inhibits DNA synthesis, leading to apoptosis in rapidly dividing cells. - **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis by incorporation into DNA and inhibition of ribonucleotide reductase. - **Hydroxychloroquine** is an antimalarial and immunomodulatory drug that also acts as an inhibitor of late-stage autophagy by preventing the fusion of autophagosomes with lysosomes. In cancer therapy, hydroxychloroquine may enhance the cytotoxic effects of chemotherapy by blocking tumor cell survival mechanisms mediated through autophagy. This combination has been investigated primarily for advanced solid tumors and small cell lung cancer (SCLC), aiming to improve outcomes over standard chemotherapy regimens. The rationale for adding hydroxychloroquine is to inhibit autophagy—a process exploited by tumor cells for survival under stress—thereby sensitizing them to chemotherapeutic agents[1][2][3][4][5].
