Overview

This is a combination regimen consisting of three agents: - **Carboplatin** is a platinum-based chemotherapeutic agent that induces DNA crosslinking and apoptosis in rapidly dividing cells. - **Gemcitabine** is a nucleoside analog (antimetabolite) that inhibits DNA synthesis, leading to cell death in proliferating tumor cells. - **Lacnotuzumab** is a humanized monoclonal antibody targeting colony-stimulating factor 1 (CSF1), thereby inhibiting the CSF1-CSF1 receptor pathway involved in the recruitment and function of tumor-associated macrophages. This combination has been studied primarily for advanced triple-negative breast cancer (TNBC) with high levels of tumor-associated macrophages. The rationale for adding lacnotuzumab to standard chemotherapy (gemcitabine and carboplatin) was to target the immunosuppressive microenvironment mediated by CSF1 signaling. In clinical trials, this triplet showed comparable antitumor activity to chemotherapy alone but with slightly poorer tolerability[1]. The development status for this specific combination appears limited due to halted enrollment from recruitment challenges.
