This is a combination therapy consisting of three agents: - **Carboplatin** (a platinum-based chemotherapeutic agent that induces DNA crosslinking and apoptosis in rapidly dividing cells) - **Gemcitabine** (a nucleoside analog antimetabolite that inhibits DNA synthesis) - **Nadunolimab** (CAN04), a monoclonal antibody targeting interleukin-1 receptor accessory protein (IL1RAP). Nadunolimab blocks IL-1α/IL-1β signaling and induces antibody-dependent cellular cytotoxicity (ADCC), counteracting the immunosuppressive tumor microenvironment and resistance to chemotherapy. It also shows potential neuroprotective effects by reducing chemotherapy-induced peripheral neuropathy. The combination is under clinical investigation for advanced cancers, particularly triple-negative breast cancer (TNBC) and pancreatic ductal adenocarcinoma (PDAC). Early-phase studies have shown promising efficacy signals in these indications[6][7][8][9].
