Overview

This drug is a combination therapy consisting of three agents used primarily in the treatment of advanced or metastatic non-small cell lung cancer (NSCLC). Carboplatin is a platinum-based chemotherapy agent that causes DNA crosslinking leading to apoptosis. Paclitaxel is a taxane chemotherapy that stabilizes microtubules, inhibiting cell division. Linifanib (ABT-869) is an orally active, potent, and selective small molecule inhibitor targeting vascular endothelial growth factor receptors (VEGFR) and platelet-derived growth factor receptors (PDGFR), which are involved in tumor angiogenesis and proliferation. The combination aims to enhance antitumor efficacy by combining cytotoxic chemotherapy with targeted inhibition of angiogenesis pathways. Clinical studies have shown this combination to be tolerable with hematologic toxicities consistent with carboplatin/paclitaxel regimens; linifanib exposure may increase when combined with these chemotherapies. This regimen has been evaluated mainly as first-line therapy for advanced nonsquamous NSCLC[1][2][5].
