Overview

This is a high-dose chemotherapy combination regimen consisting of three alkylating agents—carboplatin, melphalan, and thiotepa. The regimen is primarily used as conditioning therapy prior to autologous stem cell transplantation in patients with aggressive lymphomas (such as non-Hodgkin's lymphoma and Hodgkin's disease), metastatic breast cancer, and high-risk neuroblastoma. Each component acts by causing DNA cross-linking and damage, leading to inhibition of DNA replication and cell death in rapidly dividing cancer cells. This combination is often chosen for patients who cannot receive total body irradiation-based regimens or when an intensive cytotoxic approach is required[1][2][4].
