Overview

carboplatin + nab-paclitaxel + atezolizumab + nogapendekin alfa inbakicept is a combination chemo-immunotherapy regimen developed by ImmunityBio for the first-line treatment of advanced or metastatic non-small cell lung cancer (NSCLC). This four-drug regimen integrates standard-of-care cytotoxic chemotherapy (carboplatin and nab-paclitaxel) with the PD-L1-targeting monoclonal antibody atezolizumab and the IL-15 receptor agonist superagonist nogapendekin alfa inbakicept (NAI, also known as N-803 or Anktiva). The mechanism of action involves carboplatin-induced DNA cross-linking and nab-paclitaxel-mediated microtubule stabilization to inhibit tumor cell division, while atezolizumab blocks the PD-1/PD-L1 checkpoint to restore T-cell activity. Nogapendekin alfa inbakicept further enhances the immune response by specifically stimulating the proliferation and activation of natural killer (NK) cells and CD8+ T cells without inducing regulatory T cell expansion. This combination is being evaluated in the Phase 3 QUILT-2.023 clinical trial.
