carboplatin + nab-paclitaxel + tislelizumab

Carboplatin + nab-paclitaxel + tislelizumab is an investigational chemoimmunotherapy combination therapy. Tislelizumab is a humanized IgG4 anti-PD-1 monoclonal antibody engineered to minimize binding to Fcγ receptors on macrophages, a design intended to abrogate antibody-dependent phagocytosis and enhance T-cell mediated anti-tumor activity. Nab-paclitaxel is a nanoparticle albumin-bound formulation of paclitaxel, a taxane that stabilizes microtubules to inhibit mitosis. Carboplatin is a platinum-based chemotherapy agent that induces DNA cross-linking, leading to apoptosis. This triple combination is primarily being evaluated as a first-line treatment for advanced squamous non-small cell lung cancer (sq-NSCLC), where the RATIONALE 307 trial demonstrated significant improvements in progression-free survival compared to chemotherapy alone. It is also under investigation for other squamous cell malignancies, triple-negative breast cancer, and esophageal cancer.
