Overview

This is a combination regimen of three anticancer agents: carboplatin, olaratumab, and paclitaxel. Carboplatin is a platinum-based alkylating agent that induces DNA cross-linking and inhibits DNA synthesis and replication, leading to cell death. Paclitaxel is a taxane-class mitotic inhibitor that stabilizes microtubules, preventing their disassembly during cell division and causing G2/M phase arrest. Olaratumab is a fully human IgG1 monoclonal antibody targeting platelet-derived growth factor receptor alpha (PDGFRα), blocking ligand binding and downstream signaling involved in tumor proliferation and angiogenesis. This triple-drug regimen has been investigated in clinical trials for advanced cancers such as non-small cell lung cancer (NSCLC) and ovarian cancer[3][5][10]. The rationale for combining these drugs includes synergistic cytotoxic effects on tumor cells through complementary mechanisms of action.
