Overview

This is a combination regimen consisting of three drugs—carboplatin, paclitaxel, and talazoparib—used in the treatment of advanced solid tumors. Carboplatin and paclitaxel are cytotoxic chemotherapy agents that disrupt cell division by damaging DNA (carboplatin) and inhibiting microtubule function (paclitaxel), leading to cancer cell death. Talazoparib is a potent oral inhibitor of poly(ADP-ribose) polymerase (PARP), an enzyme involved in DNA repair; by blocking PARP activity, talazoparib prevents cancer cells from repairing DNA damage induced by chemotherapy, thereby enhancing antitumor efficacy. This triplet regimen has been investigated primarily in phase I studies for patients with advanced or metastatic solid tumors, particularly those with defects in DNA repair pathways such as BRCA mutations. The combination has shown promising antitumor activity but is associated with significant myelosuppression as the main toxicity[1][6][7][8].
