Carboplatin + paclitaxel poliglumex is a combination chemotherapy regimen consisting of the platinum-based agent carboplatin and paclitaxel poliglumex (PPX), a macromolecular conjugate of paclitaxel with poly-L-glutamic acid. Paclitaxel poliglumex is designed to improve the delivery and tolerability of paclitaxel by increasing its water solubility and eliminating the need for Cremophor EL as a solvent, thereby reducing hypersensitivity reactions[4][5]. The conjugate slowly hydrolyzes in vivo to release active paclitaxel. Both agents act as antineoplastic drugs that disrupt cell division—carboplatin forms DNA crosslinks leading to apoptosis, while released paclitaxel stabilizes microtubules and inhibits their depolymerization during mitosis[1][4]. This combination has been investigated primarily for first-line treatment of ovarian cancer and non-small cell lung cancer (NSCLC), especially in patients who are chemotherapy-naive or have poor performance status[1][5][6]. Clinical trials have shown comparable efficacy to standard regimens but with different toxicity profiles; notably, hematologic toxicity may be greater compared with standard formulations[1][6].
