Drug intelligence / Profile preview
**Carboplatin + pegylated liposomal doxorubicin** is a chemotherapy combination primarily used for the treatment of platinum-sensitive and platinum-resistant recurrent ovarian cancer, and investigated for other solid tumors and breast cancer[1][3][5][6].\nCarboplatin is a platinum-based alkylating agent that causes cross-linking of DNA, inhibiting DNA replication and transcription, resulting in cell death. Pegylated liposomal doxorubicin (PLD) is an anthracycline antibiotic encapsulated in pegylated liposomes, enhancing its pharmacokinetics, and decreases cardiotoxicity and other side effects compared to conventional doxorubicin. PLD intercalates DNA and inhibits topoisomerase II, preventing DNA replication and promoting apoptosis. The combination provides synergistic activity because the drugs act through different mechanisms and have largely non-overlapping toxicities. This regimen is considered an effective and safe alternative to other platinum-containing combinations, showing reduced rates of neurotoxicity, alopecia, and hypersensitivity relative to regimens that include paclitaxel[1][2][3][7]. Main use is first-line or subsequent therapy for recurrent ovarian cancer, especially in platinum-sensitive disease, but also explored in other solid tumors such as small cell lung cancer and triple-negative breast cancer[1][3][4][5].
