Overview

This is a combination regimen of three anticancer agents: carboplatin, sunitinib, and paclitaxel. Carboplatin is an organoplatinum alkylating agent that induces DNA cross-linking and fragmentation, leading to inhibition of DNA synthesis and cell death. Sunitinib is an oral small molecule tyrosine kinase inhibitor targeting multiple receptors including vascular endothelial growth factor receptor (VEGFR), platelet-derived growth factor receptor (PDGFR), KIT, FLT3, and RET; it inhibits tumor angiogenesis and cell proliferation. Paclitaxel is a microtubule-stabilizing agent that disrupts mitosis by promoting tubulin polymerization and inhibiting depolymerization. This combination has been investigated in phase I/II trials for advanced solid tumors such as non-small cell lung cancer (NSCLC) and triple-negative breast cancer but was associated with significant myelosuppression resulting in frequent dose delays[1][2][3][6].
