This is a combination regimen consisting of three distinct anticancer agents: - **Carboplatin** is a platinum-based chemotherapeutic that forms DNA crosslinks, inhibiting DNA synthesis and function, leading to apoptosis in rapidly dividing cells. - **Tamoxifen** is a selective estrogen receptor modulator (SERM) that acts primarily as an estrogen receptor antagonist in breast tissue, blocking the proliferative action of estrogen on mammary epithelium. - **Topotecan** is a topoisomerase I inhibitor; it stabilizes the cleavable complex between DNA and topoisomerase I, preventing religation of single-strand breaks during DNA replication and transcription, resulting in cytotoxicity especially in rapidly dividing tumor cells. This combination has been studied for use in recurrent or refractory cancers such as high-grade gliomas. The rationale for combining these agents includes potential synergistic effects by targeting different pathways involved in cancer cell survival and proliferation[1][4].
