Carboplatin + thiotepa is a high-dose chemotherapy combination regimen used primarily for the treatment of recurrent or refractory central nervous system tumors, particularly medulloblastoma, high-grade gliomas, and other solid tumors. This combination is typically administered as part of a conditioning regimen before autologous stem cell rescue (ASCR) or autologous peripheral blood stem cell transplantation (PBSCT). Both agents are alkylating agents that damage DNA in cancer cells, preventing cell division and leading to cell death. The regimen is often used when conventional treatments have failed and is frequently combined with a third agent such as etoposide or topotecan to enhance efficacy.
