Overview

This is a combination chemotherapy regimen consisting of carboxyamidotriazole (CAI), cisplatin, and vinorelbine. - **Carboxyamidotriazole (CAI)** is a small molecule calcium channel blocker that inhibits tumor cell proliferation, metastasis, and angiogenesis by blocking non-voltage-dependent calcium channels. It also inhibits inositol triphosphate synthesis and nitric oxide formation and prevents VEGF-A-induced endothelial cell proliferation[1][6][8]. - **Cisplatin** is a platinum-based chemotherapeutic agent that forms DNA crosslinks leading to apoptosis in rapidly dividing cells. - **Vinorelbine** is a vinca alkaloid that disrupts microtubule assembly during mitosis, inhibiting cancer cell division[5][7]. The combination has been studied primarily for the treatment of advanced non-small cell lung cancer (NSCLC). In clinical trials, this regimen showed improved progression-free survival compared to chemotherapy alone but with increased rates of grade 3 or higher adverse events[1][6][8].
