Overview

This is a combination cancer vaccine composed of synthetic peptides derived from carcinoembryonic antigen (CEA) and human epidermal growth factor receptor 2 (Her2/neu). Both CEA and Her2 are tumor-associated antigens overexpressed in various cancers, including colorectal, breast, gastric, pancreatic, lung, and ovarian carcinomas. The vaccine is designed to stimulate the immune system—specifically cytotoxic T lymphocytes (CTLs)—to recognize and attack tumor cells expressing these antigens. In clinical studies for advanced colorectal cancer (CRC), this combination was administered with adjuvants such as granulocyte-macrophage colony-stimulating factor (GM-CSF) and Montanide ISA-51 to enhance immunogenicity. The mechanism of action involves induction of an anti-tumor immune response targeting both CEA- and Her2-expressing cells. Clinical trials have shown the vaccine to be well tolerated with mostly mild adverse events; immune responses were detected in a majority of participants[1][3][6]. Further research is ongoing to determine its clinical benefit as monotherapy or in combination with checkpoint blockade therapy.
