Overview

This is a multi-agent chemotherapy regimen combining seven drugs: carfilzomib (a second-generation proteasome inhibitor), thalidomide (an immunomodulatory agent), dexamethasone (a synthetic glucocorticoid corticosteroid), cisplatin (a platinum-based DNA crosslinker), doxorubicin (an anthracycline topoisomerase II inhibitor and DNA intercalator), cyclophosphamide (an alkylating agent that crosslinks DNA), and etoposide (a topoisomerase II inhibitor). This combination, sometimes referred to as KTd-PACE, is used in the treatment of multiple myeloma, particularly for relapsed or refractory cases or as part of intensive regimens for high-risk disease. Each component has a distinct mechanism of action targeting different aspects of cancer cell survival and proliferation. The regimen aims to maximize anti-myeloma efficacy by combining agents with non-overlapping mechanisms.
