Drug intelligence / Profile preview
**Carmustine intracranial wafer** is a biodegradable local chemotherapy implant marketed as **Gliadel Wafer**. Each wafer contains 7.7 mg of the nitrosourea alkylating agent carmustine in a polifeprosan 20 biodegradable copolymer matrix. Implanted into the tumor-resection cavity during neurosurgery, the matrix hydrolyzes and provides sustained local release of carmustine into adjacent brain tissue. Carmustine covalently alkylates DNA and RNA, producing cytotoxic lesions and DNA cross-links that inhibit nucleic-acid replication and trigger tumor-cell death. The product is approved for newly diagnosed high-grade glioma as an adjunct to surgery and radiation and for recurrent glioblastoma as an adjunct to surgery.
