Overview

carotuximab + bevacizumab is a combination therapy of two monoclonal antibodies targeting tumor angiogenesis. **Carotuximab (TRC105)** is a chimeric IgG1 antibody that inhibits endoglin (CD105), an accessory receptor for transforming growth factor-β (TGF-β) and a key regulator of angiogenesis on proliferating endothelial cells. **Bevacizumab** is a recombinant humanized monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), preventing its interaction with VEGF receptors (VEGFR-1 and VEGFR-2), thereby inhibiting tumor blood vessel growth[1][6][9]. The combination is designed to potentiate anti-angiogenic activity by blocking two complementary angiogenic pathways (endoglin and VEGF) and has been studied primarily in patients with glioblastoma multiforme (GBM) previously refractory to bevacizumab. In clinical trials, the combination demonstrated tolerability and showed a median overall survival benefit in recurrent GBM, warranting further investigation[1][5][7].
