Overview

This is a combination therapy consisting of carotuximab (a monoclonal antibody targeting endoglin), everolimus (an mTOR inhibitor), and letrozole (an aromatase inhibitor). The combination was studied for preoperative treatment in hormone receptor-positive and HER2-negative breast cancer. Carotuximab targets endoglin, which is involved in angiogenesis, while everolimus inhibits the PI3K/AKT/mTOR pathway, and letrozole reduces estrogen production by inhibiting aromatase. Pharmacokinetic studies revealed an interaction between everolimus and carotuximab, with carotuximab accelerating the biodistribution and clearance of everolimus.
