carotuximab + pazopanib

**Carotuximab + pazopanib** is a combination therapy investigated for the treatment of advanced angiosarcoma. Carotuximab (TRC105) is a chimeric IgG1 monoclonal antibody that targets endoglin (CD105), a protein highly expressed on proliferating endothelial cells and implicated in angiogenesis. Pazopanib is an oral small molecule tyrosine kinase inhibitor that targets multiple receptors including vascular endothelial growth factor receptors (VEGFRs), platelet-derived growth factor receptors (PDGFRs), and c-KIT, thus inhibiting angiogenesis and tumor growth. The rationale for the combination is to enhance anti-angiogenic effects by concurrently inhibiting the endoglin and VEGF pathways; however, multiple phase 3 studies in advanced angiosarcoma showed that this combination did not improve progression-free or overall survival compared to pazopanib alone, and was associated with increased adverse events such as anemia and fatigue[1][3][5][7].
