CART-19 (University of Sao Paulo) is an academic, autologous CD19-directed chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of relapsed or refractory B-cell malignancies, including B-cell acute lymphoblastic leukemia (B-ALL) and B-cell non-Hodgkin lymphoma (B-NHL). The therapy involves the genetic modification of a patient's own T lymphocytes using a lentiviral vector to express a second-generation CAR. This receptor consists of a single-chain variable fragment (scFv) specific for the CD19 antigen, linked to 4-1BB (CD137) co-stimulatory and CD3-zeta signaling domains. Once infused back into the patient, these modified T cells recognize and eliminate CD19-expressing B-cells. The product is manufactured locally in Brazil at facilities such as Nutera-São Paulo and Nutera-Ribeirão Preto to increase accessibility within the Brazilian Unified Health System (SUS).
