Overview

CART-19 cells (developed by the Affiliated Hospital of the Chinese Academy of Military Medical Sciences) are an autologous chimeric antigen receptor T-cell (CAR-T) therapy designed to target the CD19 antigen. CD19 is a transmembrane protein widely expressed on the surface of B-cells and most B-cell lineage malignancies. The therapy involves the ex vivo genetic modification of a patient's T lymphocytes to express a CAR construct comprising an anti-CD19 single-chain variable fragment (scFv) linked to intracellular signaling domains (typically CD3-zeta and a co-stimulatory domain like 4-1BB or CD28). Upon re-infusion, these engineered T cells recognize and bind to CD19-positive cells, leading to T-cell activation, proliferation, and the cytotoxic elimination of the target leukemic cells. This specific product has been evaluated in Phase 1/2 clinical trials for relapsed or refractory acute lymphoblastic leukemia (ALL).
