Overview

CART-meso cells is an autologous chimeric antigen receptor (CAR) T-cell therapy developed by Shenzhen BinDeBio Ltd. for the treatment of mesothelin-expressing solid tumors, including pancreatic and stomach cancers. The therapy utilizes a second-generation CAR construct, where a patient's own T cells are lentivirally transduced to express an anti-mesothelin single-chain variable fragment (scFv) linked to 4-1BB (CD137) costimulatory and CD3ζ signaling domains. Mesothelin is a cell surface glycoprotein that is highly overexpressed in pancreatic adenocarcinoma and other malignancies but has limited expression in normal tissues, making it a viable therapeutic target. The treatment protocol typically involves lymphodepletion with cyclophosphamide prior to infusion to improve the expansion and persistence of the CAR-T cells in the tumor microenvironment.
